It's going to be a game with awful sightlines between two (currently) mediocre programs existing solely to take the record. Michigan will still have the record attendance for games people care about
To answer your question, no, a once every four years event doesn't make the sport major, ask the Winter Olympics. It is trending in the right direction but I don't think it is there yet.
I love those records and I really do not want to draw a lower crowd than 109,318 for any game this year.

Scanning some British newspapers (see links below), they seem to have been very impressed with Michigan Stadium. (Their command of geography is a bit iffy though - "Ann Arbor" and "Detroit" are used interchangeably.)

that Chicago's Soldier Field claims:
The all-time collegiate attendance record of 123,000 plus was established November 26, 1927, as Notre Dame beat the University of Southern California 7–6.
This was, of course, before the spaceship landed and made the stadium the third smallest in the NFL. And it looks like they don't have official records to give an exact attendance.
Soldier Field never seated that many. They claim to have admitted a huge number of standing-room fans that day, but can't come up with the evidence, and the NCAA doesn't accept their claim.
That Football claim will be in jeopardy in 2016. Tennessee and Virginia Tech play at Bristol Motor Speedway - capacity of 160,000.
I wonder what Dave Brandon has planned to counter this?
